Effects of indol–3-butyric acid (IBA), plant growth promoting rhizobacteria (PGPR) and carbohydrates on rooting of hardwood cutting of MM106 Apple rootstock

Abstract

This investigation was conducted to evaluate the effects of a range of indole- 3-butyric acid (IBA) concentrations (1000, 2000 and 4000 ppm), two strains of Agrobacterium rubi (A-18) and Bacillus subtilis (OSU-142) and four carbohydrates (Glucose, Sucrose, Sorbitol and Mannitol) alone, in combination with two and three treatments on the rooting capacity of the hardwood cuttings of MM106 apple rootstock in greenhouse conditions. No rooting was obtained from control treatments but only low callus formation (10%). Single treatments did not induce rooting, except IBA-1000 ppm, OSU- 142 and A-18, but induced better callus formation (20%) compared to control. Double and the three combinations were more successful in terms of rooting and callus formation. A-18+sorbitol, OSU-142+sorbitol+IBA-2000, A-18+sorbitol+IBA- 2000 and A-18+sorbitol+IBA-4000 treatments were obtained from the highest rooting formation (30%) and OSU-142+sorbitol treatment had the highest callus rate (70%), that may be a precursor of adventitious root formation. IBA-1000 ppm treatment had the highest adventitious root number (16.5). A-18 treatment had the highest average adventitious root thickness (1.61 mm). The results indicate that double and triple combination of IBA, bacteria and carbohydrates are more effective in increasing rooting capacity and more quality rooting when compared to control, or carbohydrate, IBA and, bacteria alone.

Determining the chilling requirements of four Pistachio cultivars in Semnan province (Iran)

Abstract

Most deciduous trees enter dormancy in respect to the shortening day length in fall; dormant buds re-quire a period of chilling to break this rest. The present research was carried out under laboratory con-ditions in order to determine the chilling requirements of 4 local cultivars of Damghan city. In this ex-periment some traits of flower bud breaking of mentioned cultivars in different chilling times (500, 600, 700, 800, 900, 1000, 1100, 1200, 1300, and 1400 h) were studied. So an experiment as factorial in com-pletely randomized design established in two levels of cultivar and chilling. Results indicated that the chilling requirements of Akbari cultivar were higher than others. This was due to genetic differences among cultivars. Due to the direct effect of the chilling amount during blooming period from the very entering time of the buds to the greenhouse a measure known as the Chilling Requirement Index (CRI) was used. This index seemed to calculate the chilling effects more accurately. Chilling requirements were estimated to be 1100 h for Khanjari, Shahpasand, and Abasali cultivars and 1200 h for Akbari cultivar respectively. However, chilling more than the above amount was found to be effective on re-duction of the bud blooming time.

Effect of temperature on germınatıon bıology ın Centaurea specıes

Abstract

The temperature requirement is important factor for adaptation to an ecology of any plant species.Plant seeds growing in the places with high altitutes are excepted to well develop at lower temperatures. In addition, temperature highly determines when seeds will germinate under field conditions.With different flower colors and appearence, Centaurea is an annual,biennial and perennial plant genus,alternatively used for culture plants in landscape architecture works. Centaurea virgata growing at 1750 altitude were germinated at different temperatures.Their germination rate and their percentage of germination were investigated and the best germination temperature assessed. In this study,seeds from roadsides and fields of Centaurea balsamita,Centaurea iberica and C. virgata growing at 1750 altitute were germinated at different temperatures.Their germination speed and germination percentage were investigated and the best germination temperature was assessed.As a result, the best germination percentage and speed was C. balsamita at 15°C.

Evaluation of botanical plants powders against Zabrotes subfasciatus (Boheman) (Coleoptera: Bruchidae) in stored haricot beans under laboratory condition

Abstract

Experiments were conducted to evaluate the efficacy of botanical powders of Jatropha curcas (L.), Datura stramonium (L.), Chenopodium ambrosioides(L.), Phytoloca dodecondra (L’Herit), Azadrachta indica (A. Juss) and Parthenium hysterophorus (L.) against Zabrotes subfasciatus (Boheman) in the laboratory under ambient condition. The test insects were reared in glass jar and tested on whole haricot bean grains. For comparison, primiphos-methyl and untreated check were used. The experiment was arranged in a completely randomized design in three replications. Hundred percent mortality of Z .subfasciatus was obtained with C. ambrosioides leaf powder at all levels of concentrations 24 hour after treatment application. More than 90% mortality of adult Z. subfasciatus was also observed for bean seeds treated with J. curcas, D. stramonium and P. dodecondra 96 hour after treatment at the rate of 15g/ 150g of grain application. Powder treatments of C. ambrosioides and A. indica at all tested rates and D. stramonium at higher levels (10 and 15g) gave more than 97% inhibition of F1 progeny production by Z. subfasciatus. Powder plant materials significantly reduced percent haricot bean grain infestation by Z. subfasciatus. Moreover, these plant materials had no effect on the germination capacity of haricot bean seeds. In general, the results obtained indicate that the use of these powder plant materials can be used for the control of Z. subfasciatus.

Effect of IBA concentrations on the rooting of pomegranate (punica granatum L.) cv. ganesh hardwood cuttings under mist house condition

Abstract

The experiment site was conducted under mist chamber at Horticulture Research Center, HNB Garhwal Central University, Chauras Campus Srinagar (Garhwal), Uttarakhand, India, situated in the Alaknanda valley at 300 13’ 25.26’’ N and 780 48’ 04.93’’ E and 563 m above mean sea level. The average temperature and relative humidity inside the mist house during experiment was 30+3 0C and 77+5%, respectively. The soil temperature measured was around 25+2 0C.  The hardwood cuttings of Punica granatum L. cultivar Ganesh were collected from healthy vigorous shoots of 4-6 year old plants. For preparing rooting media one part sandy soil and one part of FYM were mixed thoroughly. The stem cuttings of Punica granatum L. treated with IBA solutions of different concentrations i.e. 1, 2, 3, 4, 5g.L-1 and control by quick dip method. The experiment was replicated thrice with 10 cuttings in each treatment. A total of 180 cuttings were tested. Among all the treatments, maximum number of sprouted cuttings (7.33), average length of sprout (20.53 cm), average number of leaves (25.33), percentage of rooted cutting (73.33), number of primary roots (29.26), and average length of roots (24.88 cm) was noticed in 5g.L-1 concentration of IBA.

Effect of different substrates on survival and growth of transplanted orchids (Dendrobium nobile cv.) into net house

Abstract

Orchids are considered the most beautiful and valuable cut flower and potted plants all over the world. Although, micropropagation has become very important nowadays to meet growing market demand, there are still some barriers that hamper the ultimate goal to achieve viable ex vitro plants. Acclimatization has remained very serious issue. The current study was carried out to investigate the impact of different substrates available in nature on the survivability and development of ex vitro plantlets under net house conditions. The step-wise protocol of acclimatization utilized plantlets with well-developed roots produced in MS rooting medium involved NAA at 3.0 mg l-1. The substrates used for acclimatization were different mixtures of coco-peat, desert sand, bricks chips, saw dust, coal pieces and date palm chips. The treatment contained coco-peat: desert sand: perlite at ratio 1.0:1.0:0.2 (v/v/v) proved the best mixture with 100% survival rate after 120 days of transplantation. Furthermore, addition of wood and bark chips to this best soil substrate as an upper layer improved the subsequent growth and development of plants. Number of new shoots and other growth parameters were recorded and the whole protocol is discussed in the current study.
