Biodesulfurization of dibenzothiophene and its alkylated derivatives through the sulfur-specific pathway by the bacterium RIPI-S81

Abstract

RIPI-S81 is a new dibenzothiophene (DBT)-desulfurizing bacterium, which was isolated by Research Institute of Petroleum Industry in Iran. Resting cells and growing cells of RIPI-S81 was able to convert alkylated dibenzothiophenes (Cx-DBTs) to hydroxybiphenyls such that they were almost stoichiometrically accumulated as the dead-end metabolites of Cx-DBTs desulfurization in the medium containing minimal salt (MSM) and nutrients. RIPI-S81 could desulfurize up to 80% of 4,6-dimethyldibenzothiophene and 50% of methyldibenzothiophene in the MSM containing 40 mg/l of a sulfur source. The molecular structures of metabolites and the reduction of Cx-DBTs were analyzed using GC-MS and HPLC. The position of alkyl substitutes and the sulfur substrate affected desulfurization rates.

In vitro production of growth regulators and phosphatase activity by phosphate solubilizing bacteria

Abstract

Crops absorb phosphorous in the form of soluble orthophosphate ion. The solubility of phosphate is inhibited by the presence of iron and aluminium in acidic soils and calcium in neutral and alkaline soils. This leads to fixation of phosphorous, making it not available to crop plants. The phosphate solubilizing bacteria (phosphobacteria) secretes some kinds of organic acids which act on insoluble phosphates and convert the same into soluble form, thus providing phosphorous to plant. An experiment was conducted to enumerate the population density of phosphobacteria in the rhizosphere soils of brinjal, chilly, cotton, green grain, groundnut, maize, paddy, ragi, sorghum and turmeric using Ketznelson and Bose medium following dilution plate technique. Efforts have been made to isolate phosphobacteria from these soils and isolated strains were inoculated in specific media containing specific substrates to produce growth regulating substances such as IAA and GA3 and phosphatase enzyme. The result showed that the population levels of phosphobacteria were higher in the rhizosphere soil of groundnut plant. Further, all the strains of phosphobacteria were able to produce phytohormones and phosphatase enzyme under in vitro conditions.

Bacteria associated with compost used for cultivation of Nigerian edible mushrooms Pleurotus tuber-regium (Fr.) Singer, and Lentinus squarrosulus (Berk.)

Abstract

The bacteria involved in an outdoor single phase composting using sawdust and wheat bran as substrates for cultivation of Pleurotus tuber-regium (Fr.) Singer, and Lentinus squarrosulus (Berk.), two Nigerian edible mushrooms were identified. Composting was carried out for 2 weeks. The highest core and peripheral temperatures were 68 and 48ºC, respectively, while the lowest temperature was 32ºC. The highest number of bacteria in the core and peripheral compost were 1.46 x 106 and 6.90 x 105 cfu/ml, respectively. Bacteria isolated and characterized from the fermenting agricultural substrates include Bacillus polymyxa, Enterobacter aerogenes, Micrococcus roseus, Citrobacter freundii, Bacillus subtilis, Clostridium perfringens, Bacillus licheniformis, Pseudomonas aeruginosa, Bacillus cereus and Escherichia coli. The implication of the presence of these bacteria is discussed.

Effects of application of phosphorus fertilizer on brown blotch disease of cowpea

Abstract

A field experiment was conducted to investigate the effects of level of phosphorus application on brown blotch disease of cowpea during the planting seasons of 2001, 2002 and 2003. Application of phosphorus from Single Super Phosphate at 30, 60, 90, and 120 kg P2O5/ha significantly increased the number of petioles, pods, nodules, seed/pod leaf area and yield. The higher the level of application of phosphorus the higher values of all the parameters. Disease incidence and severity of brown blotch were significantly reduced at higher levels of phosphorus (90 and 120 kg/ha) irrespective of the method of application. Moreover, there were also increases in the grain yield of cowpea

Expression of melanin and insecticidal protein from Rhodotorula glutinis in Escherichia coli

Abstract

Both the salmon/red melanin and the insecticidal producing genes of Rhodotorula glutinis was successfully expressed in Escherichia coli using plasmid pZErO -1. This work suggests that in Rhodotorula species melanin and insecticidal toxin are co-expressed and therefore possibly co-evolved.

Subcloning and expression of human alpha-fetoprotein gene in Pichia pastoris

Abstract

Alpha-fetoprotein protein (AFP) is naturally found in fetal serum while production and appearance after birth is indicative of presence of malignant tumors. Therefore, by measuring this protein during fetal period and after birth, it is possible to diagnose abnormalities and tumors in fetus or the newborn. The objective of this study was to produce and purify AFP protein using DNA recombinant technology to apply in diagnostic kit preparations. In this study Pichia pastoris as metylotrophic yeast was used for AFP production. After construction of recombinant plasmid, pS1-AFP, electroporation and lithium chloride techniques were used for transferring to susceptible cells. The quantity and quality of the produced protein were checked by SDS-PAGE and ELISA methods. Selection of transformed mutant strains of Muts and culturing them in glycerol media (YPG) up to OD600=6 and their transfer to methanol media (YPM) with augmentation of methanol to 1% final concentration resulted in inducing protein production in auxotrophic media lacking histidine. This protein could be useful in monoclonal antibody production and in diagnostic kit preparations.
