Cytotoxic Effects of Angelica decursiva on Osteosarcoma Cells: Induction of Apoptosis and Inhibition of Proliferation

The Angelica decursiva has been used in Korean traditional medicine as an antitussive, an analgesic, an antipyretic and a cough remedy. However, its anticancer activity has been unknown. In this study, we have examined the cytotoxic activity of ethanol extract of A. decursiva root (EEAD), and the mechanism by which EEAD suppresses cell growth in Saos2 human osteogenic sarcoma cells. Treat-ment of EEAD in Saos2 cells induced the apoptic cell death in a concentration-dependent manner as determined by MTT assay and DNA fragmentation analysis but not in FOB human normal osteoblast cells. Furthermore, the proteolytic processing and the activity of caspase-3 and -7 were increased by EEAD treatment in Saos2 cells. These results suggest that EEAD can induce the suppression of cell growth and cell apoptosis in Saos2 human osteogenic sarcoma cells, and therefore, it may have potential properties for anticancer drug discovery.

Antimicrobial Activity of Allium sativum Linn. Extracts Against Hospital-Acquired Bacteria

The presence of pathogenic microorganisms causing infections to hospitalized populations in the 750 bed Specialist Hospital, Yola, Nigeria was investigated for a period of two and a half years. Staphylococcus aureus, Escherichia coli, Streptococcus pneumoniea and Pseudomonas aeruginosa were selected through the use of questionnaires as the commonest bacteria causing nosocomial infections, and were isolated from clinical specimens obtained from patients who were admitted to the hospital for reasons other than the infections caused by these bacteria. The effects of water, ethanol and chloroform extracts of garlic against the nosocomial S. aureus, E. coli, S. pneumoniea and P. aeruginosa were investigated under various use conditions, such as variations in temperatures and pH. At a concentration of 100 mg/ml, all the crude extracts inhibited the growth of the pathogenic bacteria, though with varying degrees of susceptibility. However, laboratory based bacteria which were used as controls were more susceptible than their corresponding nosocomial bacteria. Activities were stronger under elevated temperatures and slightly acidic pH values. The MBC values of the aqueous extract for S. aureus was 75 mg/ml; S. pneumoneae, 100 mg/ml; E. coli, 125 mg/ml and P. aeruginosa, 150 mg/ml. The water extract was more potent than the organic extracts, and all were inferior in activity, when compared to the standard antibiotic, metronidazole. The gram positive S. aureus was more susceptible to the toxic effects of garlic than its gram negative counterparts. The results obtained in this study indicate that water extracts of garlic can be used alongside conventional antibiotics to fight agents of nosocomial infections that are so prevalent in our hospitals.

Pharmacological Assessment of Pain Relief from Datura metel Seed Extract

This work was carried out to investigate the analgesic activity of dried seed of Datura metel Linn. A concentrated aqueous extract was obtained, mimicking conditions used in traditional treatment setting, which includes dissolving the extract in water. As in this part of the world, experiments were carried out with the aqueous extract for its peripheral and central antinociceptive potentials on acetic acid- induced writhing and radiant heat tail- flick models in rats, respectively. There was sensation of pain by the rats administered with the extract in the two test models used, after 60 min and above of pretreatment with the seed extract of D. metel. Hence, the analgesic activity of D. metel seed extract was found not to be significant ( P>0.05) on acetic acid induced model, as well as the radiant heat tail-flick model. The behavioural pattern of sedation and decreased appetite on administration of seed extract could be explained on the basis of the action of some receptors like µ- receptors in the CNS, which when stimulated have the intrinsic potential to reduce the distress or the affective component of pain without having any significant change in the intensity of the actual sensation.

Antimycotoxigenic Compounds from Warburgia salutaris: Isolation of Active Furans and Furanones

Warburgia is also known as Pepperbark, Isibhaha and Peperbasboom. This tree is one of most valuable of the natural African antimicrobials and the stem and leaves have been used to treat yeast, fungal, bacterial and protozoal infections for centuries. The bark of Warburgia salutaris is used in traditional medicine as an expectorant and smoked for coughs and colds. It is also used as a topical application for sores and inflammation. Previous screening of W. salutaris showed that this plant had promising antibacterial activity. Subsequently, this endangered tree species was selected for bioassay-guided purification in order to identify the active principles. Fractionation of the toluene: ethyl acetate (7:93 v/v) extract of the stem bark by chromatographic techniques yielded a sesquiterpenoid which exhibited antimicrobial activity against Gram-positive bacteria. The compound, muzigadial, has previously been reported in two other Warburgia species, this being the first time (5A , 9A , 9B )-5, 5A, 6, 7, 8, 9, 9A, 9B-Octahydro-6, 6, 9A-trimethylnaphtho[1, 2c] furan-1-(3H)-one (drimenin) and 5, 10-Dihydro-6, 7-dimethyl-4H-benzo [5, 6] cyclohepta [1, 2-b]-furan has been reported from W. salutaris.

Pharmacological Effects of Methanolic Quercus infectoria Bark Extract on Lipid Profiles, Blood Glucose, Gastric Ulceration, and Antimicrobial Activity

Nowadays, the surge of consumption of herbal supplements is encouraged by several factors, including the common belief that all herbal products are relatively safe and effective. The present investigation explores the effects of methanolic extract of Quercus infectoria bark upon rat blood lipid profile, glycemia, inflammation, gastric ulcer and bacterial growth. After one month of chronic extract (0.5% w/v) intake via drinking water, there was a significant increase in serum HDL-cholesterol level. This was accompanied with an increase in both serum glucose and insulin levels. No significant changes were observed in other lipid parameters studied. Liver enzyme activities as well as urea and creatinine levels were not negatively affected. Extract at 250, 500 and 1000 mg/kg body weight exhibited substantial anti- inflammatory effects in cases of acute and chronic inflammation induced by carrageenan and formalin respectively. Pre-treatment of fasted rats with the extract (100 and 500 mg/kg body weight) also demonstrated significant protection against ethanol-induced gastric ulcer. Anti-bacterial activity against Proteus mirabilis, Citrobacter braaki, and Staphylococcus aureus methicillin resistant and sensitive was also noticed. In conclusion, these findings suggest that the methanolic extract of Q. infectoria bark provides an inexpensive and powerful source of herbal supplement used to treat various conditions.

Inhibitory Effects of Ficus hispida on Breast Cancer Cell Proliferation In Vitro

Stems of Ficus hispida L. have long been prescribed as one of the constituents in various Thai traditional remedies for cancer therapy. In the present study, crude ethanol extract and its sequential fractions from F. hispida L.: water, methanol: water, methanol and ethyl acetate fraction were tested in vitro against SKBR3, MDA-MB435, MCF7 and T47D human breast cancer cell lines. The results have shown that the methanol extract exhibited antineoplastic activity against T47D cells. The cytotoxic activity was further examined by MTT assay with more dilution, colony forming assay and cell cycle analysis. The IC50 of this extract against T47D cell was 110.3 +/-9.63 g/mL by MTT assay and colony forming assay confirmed the cell growth inhibition in a dose-dependent manner. Cell cycle analysis demonstrated a rising of apoptotic cell population in herbal treated cells. Therefore, F. hispida L. used in traditional medicine may provide some benefits in the treatment of breast cancer.
